This application note describes a method for analyzing oxycodone and its metabolites (noroxycodone, oxymorphone, and noroxymorphone) in plasma using high-performance liquid chromatography/mass spectrometry (HPLC/MS) and the Agilent ZORBAX Rapid Resolution High Throughput (RRHT) StableBond SB-C18 column for chromatographic separation. The method employs an ammonium acetate/acetonitrile gradient and detection by electrospray positive ionization mode mass spectrometry. Spiked human plasma samples are subjected to solid-phase extraction prior to HPLC/MS analysis. The method provides fast, efficient analysis with minimal solvent consumption, which increases productivity, and good linearity (R2 > 0.9900) and reproducibility (less than 10% difference between replicate analyses) for all compounds.

The Agilent Cary 60 is a UV-Vis spectrophotometer produced by Agilent. It uses a classic Cary optical design with a Czerny-Turner monochromator. The spectral range is 190-1100nm, the resolution is less than 1.5nm, and a single xenon light source is used with a lifetime of up to 10 years. It has two silicon detectors, multiple layers of quartz optical elements, a scanning speed of up to 24000nm/min, a maximum speed of 80 points/second for time events, and a grid shift in non-measurement. It is immune to external scattered light and is fully controlled by a PC via Windows and a USB port. It has GLP and CFR 21 Part 11 software for complete instrument validation in accordance with all pharmacopoeias.
