This application note describes a method for analyzing oxycodone and its metabolites (noroxycodone, oxymorphone, and noroxymorphone) in plasma using high-performance liquid chromatography/mass spectrometry (HPLC/MS) and the Agilent ZORBAX Rapid Resolution High Throughput (RRHT) StableBond SB-C18 column for chromatographic separation. The method employs an ammonium acetate/acetonitrile gradient and detection by electrospray positive ionization mode mass spectrometry. Spiked human plasma samples are subjected to solid-phase extraction prior to HPLC/MS analysis. The method provides fast, efficient analysis with minimal solvent consumption, which increases productivity, and good linearity (R2 > 0.9900) and reproducibility (less than 10% difference between replicate analyses) for all compounds.
